Is it possible to have too many one way links from a single website? ie is there a trigger point when the Search Engines start to ignore new links?

Yes it is possible to have many links but the links should be various sources.

Generally websites/directories do not link to the same site more than once. However, It is possible to get one way links from a single website if it is social networking site / community website. But it is not beneficial as Google will take it just one vote in favor of your site no matter how many one way links you have from there.

Either you should submitting the site in variety of directories and in the relevant categories only for sure. You can start with this list of 17000 web directories

Yes it is possible but be sure that all of your one way links are good quality links.
