On page optimization is the fundamental of SEO but known to be the complicated part. It is having by making changes, optimizing and bringing the best structure in your site.
Off page optimization is absolutely getting links in your page, the possible things you could get to make a high Page rank through outside your website.

On page optimization - deal with the website content, analysis, competitors, design, link placing and keyword researching. In layman's term, more about your website's inner optimization.
Off page optimization - all about link building like link submission, forum participation, blog comment, classified posting etc. In layman's term, outside website link promotion.

On site SEO is the practice of optimising your content, meta data, keyword density etc and off page optimisation are things like gaining links from other websites, publishing PR articles, viral marketing etc etc

On-page optimization includes content management, finding the right keywords, may factors that will be considered on your website.  While off-page optimization includes building links on other websites by posting your url.  Link building includes several methods like blog commenting, social bookmarking, forum posting, article submission, directory submissions and many more.
