I just had my first IUI and was on clomid. I have started spotting and today was my two weeks after test. I took a pregnancy test it said negative. The Dr. wants me to take another on Friday. What does this mean if I am spotting?

Unfortunately, if your test was negative and you are spotting, it is likely that you're getting your period and that this cycle was not successful.
The test on Friday will give you the final answer regarding the success of the cycle.
